【摘要】 主要進(jìn)行雙孢蘑菇菌種液氮保藏中菌絲的培養(yǎng)基質(zhì)、不同冷凍保護(hù)劑、不同降溫速率及菌種解凍方式等因素對(duì)菌種活力的影響研究。結(jié)果表明生長(zhǎng)3 d5 d菌齡的PDA菌絲塊,以10%甘油作為冷凍保護(hù)劑及聚丙烯吸管安瓿為容器,制備的菌種保藏管在液氮罐口緩慢降溫至-40℃左右時(shí),迅速浸入液氮保藏,提取的液氮保藏菌種以快速解凍方式進(jìn)行復(fù)蘇,雙孢蘑菇菌種經(jīng)過4年的液氮保藏,存活率達(dá)100%,栽培出菇的農(nóng)藝性狀仍保持原有菌株特性,研究建立的雙孢蘑菇液氮保藏方法已成為我所蘑菇菌種長(zhǎng)期保藏的主要方法之一。
【Abstract】 To investigate the optimum conditions of liquid nitrogen preservation of Agaricus bisporus spawn stocks,four preservation factors,i.e.culture substrate of mycelium,cryoprotectants,cooling rate and thawing methods were discussed in this study.The results showed that PDA medium blocks after 3 d5 d,10% glycerol as cryoprotectant and Polypropylene-straw-ampoules as container were the best.In cooling process,strain tubes for preservation immersed into liquid nitrogen jar rapidly after cool...
