【摘要】 以蝙蝠蛾擬青霉(Paecilomyces hepiali)菌絲體為材料,以腺苷和D-甘露醇為評(píng)價(jià)指標(biāo),在單一酶提取實(shí)驗(yàn)基礎(chǔ)上,采用正交實(shí)驗(yàn)設(shè)計(jì)對(duì)復(fù)合酶法提取工藝進(jìn)行優(yōu)化。優(yōu)化的復(fù)合酶法提取工藝:浸提pH 5.5、浸提溫度40℃、酶組合為纖維素酶0.1%+蝸牛酶0.1%,在此工藝條件下,提取物中甘露醇、腺苷含量分別為12.56%和2 065μg/g。
【Abstract】 Factors affecting the enzymatic extraction of D-mannitol and adenosine from Paecilomyces hepiali mycelium were optimized using orthogonal tests designed on the basis of single factor experiments.The data revealed that the optimal extraction pH,temperature and enzyme combination values were pH 5.5,40 ℃,and 0.1% cellulase and 0.1% snailase,respectively.Maximal levels of mannitol and adenosine obtained under optimal extraction conditions were 12.56% and 2065 μg/g,respectively.
