【摘要】 利用DEAE-Sepharose Fast Flow離子柱層析和Sephacryl凝膠柱層析對灰樹花(Grifola frondosa)子實體水提醇沉粗多糖進行分離純化,對分離純化到的一種多糖GFP75-2-2B進行了分析。紫外光譜鑒定結(jié)果表明,GFP75-2-2B不含蛋白質(zhì)、核酸和酚類成分;離子色譜分析表明其單糖組成為巖藻糖、鼠李糖、半乳糖、葡萄糖和甘露糖,摩爾比為1.0∶1.3∶3.6∶4.0∶4.5。GFP75-2-2B對巨噬細胞株RAW264.7進行體外免疫活性的研究結(jié)果表明,GFP75-2-2B有明顯刺激巨噬細胞分泌NO的活性。
【Abstract】 A water-soluble,crude polysaccharide preparation was obtained from Grifola frondosa fruit bodies by hot water(100 ℃) extraction followed by ethanol(75%) precipitation.A purified polysaccharide,GFP75-2-2B,was obtained from the crude material by successive fractionation using DEAE-Sepharose Fast Flow ion exchange chromatography and Sephacryl S gel filtration chromatography.UV spectroscopy detected no protein or nucleic acid in GFP75-2-2B,which consisted of fucose,rhamnose,galactose,glucose and man...
