【摘要】 采用組織分離法,對(duì)東寧縣的松口蘑進(jìn)行了分離培養(yǎng)。通過顯微觀察及RAPD-PCR技術(shù)對(duì)松口蘑子實(shí)體及其相應(yīng)的培養(yǎng)物之間進(jìn)行了親緣關(guān)系鑒定。結(jié)果表明,菌褶為松口蘑組織分離的最適宜部位,其最佳分離培養(yǎng)基為PWJ,即PDA+黑木耳二級(jí)種浸出液+麥麩。RAPD-PCR結(jié)果顯示,松口蘑子實(shí)體與分離物之間的DNA指紋相似系數(shù)在0.97~1.00之間,表明松口蘑子實(shí)體與其相應(yīng)的培養(yǎng)物在種質(zhì)上是一致的。 更多還原
【Abstract】 In this study,the mycelia of Tricholoma matsutake from Dongning were cultivated by the method of tissue separation and identified using RAPD-PCR.The results showed that lamellae were suitable for the tissue separation and well induced on PWJ medium.By RAPD-PCR analysis,DNA fingerprint similarity coefficient was 0.97~1.00,that implied the mycelia were indeed from their corresponding fruiting body. 更多還原
