【摘要】 篩選出2株昭通本地天麻共生蜜環(huán)菌(SNA03、SNA04)菌索生長(zhǎng)的最佳條件。采用單因素試驗(yàn)和正交試驗(yàn)法進(jìn)行篩選,以生物量為考察指標(biāo)。2株菌菌索生長(zhǎng)的最適溫度為25℃,暗培養(yǎng)下菌索生長(zhǎng)速度最快;單糖(葡萄糖)為2菌株生長(zhǎng)的最佳碳源,有機(jī)氮中的酵母膏和蛋白胨為最佳氮源,最適無(wú)機(jī)元素為K2SO4(SNA03)和KH2PO4(SNA04),最適維生素為VB2;最佳營(yíng)養(yǎng)條件組合為葡萄糖∶酵母膏∶K2SO4(SNA03)或KH2PO4(SNA04)∶VB2=15g·L-1∶3g·L-1∶2g·L-1∶0.005g·L-)1,氮源(酵母膏)是影響菌索生長(zhǎng)的最主要因素。來(lái)自昭通的2株天麻共生蜜環(huán)菌對(duì)營(yíng)養(yǎng)和環(huán)境條件的要求基本一致。最佳生長(zhǎng)條件的確定,對(duì)昭通蜜環(huán)菌快速工業(yè)化制種及天麻的大規(guī)模生產(chǎn)將起到一定的推動(dòng)作用。
【Abstract】 The best conditions for rhizomorph growth of 2 Armillaria mallea strains (SNA03, SNA04) which were symbiotic with Gastrodia elata in Zhaotong prefecture of Yunnan province were screened out. The best growth conditions were determined by single-screening and orthogonal test and the biomass was regarded as the investigated index. 25℃ was the optional temperature and the growth rate were most quickly under dark cultivation. Monosaccharide (glucose) and organic nitrogen (yeast extracts, protein peptone) were the optional carbon source and nitrogen sources separately, inorganic salt (potassium for SNA03, potassium phosphate for SNA04) were more favorable for rhizomorph growth than others and VB2 was the best growth factor. Nitrogen source (yeast extract) was the most important factor for rhizomorph growth. The requirements for nutrient and environmental conditions of 2 strains were basically the same.
