【摘要】 研究北蟲草超氧化物歧化酶(SOD)的分離純化和部分酶學特性。結(jié)果表明,從北蟲草中提取SOD的最佳工藝條件為原料與PBS(pH7.73)配比為1∶3(m∶v),提取時間為3h,純化倍數(shù)可達到粗酶液的4.09倍。提純后的SOD在690nm左右出現(xiàn)了特征吸收峰,最適溫度38℃,最適pH值為7.8,大部分糖對SOD的活性沒有顯著影響,有機酸都有降低酶活性的作用,過氧化氫對該酶有抑制作用,北蟲草SOD是以銅和鋅離子為輔基,為北蟲草的進一步開發(fā)和利用提供了理論依據(jù)。
【Abstract】 The properties of superoxide dismutase (SOD) from Cordyceps militaris were studied. The optimal isolating conditions were that the ratio of crude extract and PBS was 1∶3 (m∶v) , the pH value of PBS was 7.73, and the extraction time was 3 h. The purification multiple was 4.09 by column chromatography. The purified SOD showed characteristic peak at 690 nm. The optimum temperature and pH were 38℃ and 7.8, respectively. Lactose, sucrose, glucose and maltose had no impact on the activity of SOD. The activity of SOD was inhibited by organic acid and H2O2. SOD of C. militaris was Cu, Zn-SOD. SOD from C. militaris were isolated and purified successfully, and its partial enzymatic properties were studied, which provided important information for future applications and research.
