【摘要】 從NCBI中下載了12 184條香菇(Lentinula edodes)表達(dá)序列標(biāo)簽(expressed sequence tag,EST)序列,處理后得到全長(zhǎng)3 681 177 bp的4 684個(gè)Unigene。在其中共發(fā)掘出142個(gè)簡(jiǎn)單重復(fù)序列(simple sequencerepeat,SSR),分布于120條EST中,分布頻率為2.99%(平均分布距離:25.924 kb)。其中,單、二、三核苷酸重復(fù)是主要的重復(fù)類型,A/T,AG/CT,ACG/CTG是單、二、三核苷酸的主要重復(fù)基序,分別占所有EST-SSR的23.23%、21.83%和10.56%。利用引物設(shè)計(jì)軟件PrimerPremier5.0設(shè)計(jì)了40對(duì)引物,并利用6%變性聚丙烯酰胺凝膠在18個(gè)香菇品種中進(jìn)行檢測(cè)。其中,22對(duì)引物為多態(tài)性引物,多態(tài)率為55%。應(yīng)用NTSYS軟件的聚類方法分析表明,18份材料遺傳相似系數(shù)范圍為0.3750.984,與前人研究結(jié)果相符。
【Abstract】 A total of 12 184 Lentinula edodes expressed sequence tags(ESTs) were downloaded from NCBI and shown to be associated with 4 684 Unigenes comprising 3 681 177 bp in length.A MISA search revealed a total of 142 simple sequence repeats(SSRs) with a frequency of 2.99% and an average distance distribution of 25.9 kb.The most abundant mononucleotide,dinuclotide and trinucleotide repeats were A/T,AG/CT and ACG/CTG,accounting for 23.23%,21.83% and 10.56% of each type,respectively.Forty EST-SSR primer pairs were designed by PrimerPremier 5.0,22 of which revealed polymorphic bands in the DNA of 18 L.edodes strains.NTSYS-computed similarity coefficients for the 18 strains ranged between 0.3750.984,which was approximately in accordance with their diversity based on previous research.
