【中文摘要】 通過(guò)巢式簡(jiǎn)并PCR和基因組步行技術(shù)從香菇(Lentinula edodes)單核體中克隆到乳清酸核苷-5′-單磷酸脫羧酶(orotidine-5’-monophosphate decarboxylase,OMPDC)的編碼基因le-pyrG,長(zhǎng)度為946bp。經(jīng)生物信息學(xué)分析,香菇le-pyrG基因含有2個(gè)長(zhǎng)度分別為67bp和51bp的內(nèi)含子,該基因可以編碼1個(gè)含有275個(gè)氨基酸殘基的蛋白質(zhì)序列,該序列經(jīng)比對(duì)分析顯示與裂褶菌(Schizophyllum commne)和灰蓋鬼傘(Coprinus cinereus)OMPDC序列的相同性分別為73%和70%,相似性分別為84%和83%。這是首次報(bào)道從栽培食用真菌中克隆出乳清酸核苷-5′-單磷酸脫羧酶編碼基因的全序列。
【英文摘要】 Le-pyrG, the gene encoding orotidine-5’-monophosphate decarboxylase (OMPDC) in Lentinula edodes, was cloned by combining the methods of nested PCR and chromosome walking. Le-pyrG is 946 bp in size, contains two introns of 67 bp and 51 bp, and the deduced protein sequence consist of 275 amino acids. Alignment analysis revealed that the deduced amino acid sequence encoded by le-pyrG exhibited 73% and 70% identity, and 84% and 83% positivity, with OMPDC enzymes reported in Schizophyllum commune and Coprinus cinereus, respectively. To our knowledge, this is the first report describing the cloning of a complete pyrG gene sequence from a cultivated edible fungus.
