【中文摘要】 優(yōu)化了以桑葉和麩皮為主要原料由毛云芝菌(Coriolus hirsutus)生產(chǎn)漆酶的培養(yǎng)條件,即培養(yǎng)料起始pH、裝料量、封口情況、接種量、發(fā)酵時(shí)間對(duì)漆酶活性的影響,并觀察了該菌在優(yōu)化后培養(yǎng)條件下的代謝動(dòng)力學(xué)情況。結(jié)果表明,自然pH,裝量為10 g/250 mL三角瓶、封口紗布4層,接種量為5 mL種子液/瓶,培養(yǎng)時(shí)間為10 d,酶活達(dá)到1.37×104U,優(yōu)化后結(jié)果比初始設(shè)計(jì)提高了18%。
【英文摘要】 Substrate composition,initial pH,ratio of substrate to cultivation vessel volume,aeration,inoculum size and cultivation time were optimized for maximal laccase production by Coriolus hirsutus grown under solid-state conditions using mulberry leaves and wheat bran as the main components of the cultivation substrate.Highest laccase levels(1.37 × 104 U per) were recorded after ten days in 250 mL culture flasks containing 10 g substrate at pH 6,sealed with four layers of cheese cloth and inoculated with 5 mL of seed culture.
