【中文摘要】 異硫氰酸熒光素(FITC)標記的紫孢側耳單核原生質體與未標記的糙皮側耳單核原生質體經(jīng)PEG融合后,在倒置顯微鏡下,用顯微操縱儀直接吸取一方帶有熒光另一方不帶熒光的原生質體對,經(jīng)培養(yǎng)后,根據(jù)“鎖狀聯(lián)合”這一形態(tài)特征挑選出融合子,融合率為5.2×10-3.進一步鑒定結果表明,融合菌株的酯酶同工酶譜發(fā)生了明顯的變化,其生物量和生長速度有不同程度的提高,子實體形態(tài)也與雙親有所不同.
【英文摘要】 Fluorescein isothiocyanate (FITC), which was not toxic to protoplasts, was used for the fusion of protoplasts. After the fusion of the FITC-labelled monokaryon protoplasts from Pleurotus sapidus and the non-FITC-labelled monokaryon protoplasts from Pleurotus ostreatus, a pair of sticked protoplasts was obtained with micromanipulator under an inverted microscope. Cultured with liquid regeneration medium, the fusants were selected according to the clamp connection of dikaryocyte with a fusion rate of 5.2?03. It was found that the estcrase isozyme zymogram of fusants changed greatly, while the biomass and growth rate of many fusants were superior to those of their parents, and that the fruitbody morphology of the fusants was different from their parents.
