【中文摘要】 長裙竹蓀(Dictyophora indusiata)子實體的3%三氯乙酸提取液經(jīng)乙醇沉淀,去蛋白后得到多糖粗制品。粗制品再經(jīng)二次DEAE-纖維素柱層析分離和Sephadex G-200柱純化得到竹蓀水溶性多糖DiA。經(jīng)Sepharose 4B柱層析、聚丙烯酰胺凝膠電泳和醋酸纖維薄膜電泳鑒定,DiA為—單一物質(zhì)。紙層析和氣相層析表明,DiA的單糖組成為葡萄糖和甘露糖,其摩爾比為:0.29:1.00。多糖的分子量約為168000。
【英文摘要】 A soluble polysaccharide DiA was obtained through being extracted from dried fruitbodies of Dictyophora indusiata by 3% trichloreacetic acid, precipitated by cthanol, deproteinized by protease method and Sevag method, then isolated twice by DEAE-cellulose column chromatography and further puried by Sephadcx G-200 column. DiA was proved homogeneous by chromatography on Sepharose 4B column, acetate cellulose membrane electrophoresis and polyacrylamide gel electrophoresis. Identified by paper chromatography and gas chromatography, it was proved that the components of DiA were composed of D-Glc and D-Man with a molar ratio of 0.29:1.00 and that its molecular weight was estimated to be 168000.
