【中文摘要】 本文報(bào)道了應(yīng)用RFLPs和RAPD技術(shù)分析香菇類緣關(guān)系,鑒別品系及分析木耳雜交后代和原生質(zhì)體融合子的結(jié)果。發(fā)現(xiàn)供試的32個(gè)香菇菌株的總DNA限制性內(nèi)切酶片段長度存在多態(tài)性;木耳雜交后代、融合子及其親株間DNA PCR擴(kuò)增產(chǎn)物譜帶型出現(xiàn)差別。這充分說明RFLPs和RAPD技術(shù)在分析遺傳變異、鑒別品系、鑒定雜種和融合子研究中的重要作用。
【英文摘要】 The analysis of Lentinus edodes phylogenetic relationship among strains, the identification of L. edodes -biological strains, the identification of interspecific protoplast fusants and hybrids of Auricularia sp. using RFLPs (Restriction Fragment Length Polymorphisms) and RAPD (Random Amplified Polymorphic DNA) techniques were reported in this paper. The results indicated that the polymorphisms of restriction fragment or genomic DNA were detected from 32 Lentinus, edodes strains tested. The electrophoretogram of PCR (Polymerase Chain Reaction) amplified products was different between fusants, hybrids and their parental straims. So it was suggested that PFLPs and RAPD techniques were much useful genetic markers in analysis of hereditary variation and identification of strains, fusants and hybrids.
