【中文摘要】 本文采用高效陰離子交換色譜—脈沖安培檢測器(HAPEC-PAD),建立了一種測定多糖中單糖比例的方法。以NaOH為淋洗液,CarboPacTMA20預(yù)處理柱,CarboPacTMPA20分離柱,金工作電極,Ag/AgCl參比電極,8種自然界中常見單糖標(biāo)準(zhǔn)品做混合標(biāo)樣,探索方法可行性。在淋洗液濃度為2.5 mmol/L時,各種單糖組分得到有效分離,其線性和重現(xiàn)性均良好。并在此基礎(chǔ)上測定了毛頭鬼傘多糖中單糖比例;與傳統(tǒng)的方法相比,此方法具有前處理簡單、靈敏度高、節(jié)省時間和試劑等優(yōu)點(diǎn)。
【英文摘要】 This paper offers a method to determine the monosaccharides'composition by high performance anion exchange chromatography-pulsed ampere detector(HPAEC-PAD).With NaOH eluent,a gold work electrode and a Ag/AgCl reference electrode,CarboPac20 PA20 guard column,CarboPacTM PA20 analytical column and 8 monosaccharides as a mixed standard the feasibility is studied.The result shows that at 2.5 mmol/L NaOH each monosaccharide can be well separated;Their linear relationship and repeatability are good.In the same condition,the monosaccharides'composition of the polysaccharides from the fruiting bodies of coprinus comatus was determined.This method is better than traditional methods because it does not need derivation reaction and detects lower concentration and saves time and reagents.
