【中文摘要】 以黑木和毛木耳為親本菌株 ,通過滅活原生質體融合 ,經(jīng)過初篩和復篩 ,一株新的穩(wěn)定的木耳高產(chǎn)菌株被選育出來。黑木耳和毛木耳原生質體分別被熱滅活 (6 5℃ ,30min)和紫外線滅活 (30W ,30cm ,10min) ,雙親存活率為 3 5× 10 -7~ 3 2× 10 -8。在聚乙二醇誘導下融合繼續(xù)生存 ,親本原生質體融合被實現(xiàn) ,重組頻率為4 3× 10 -5。
【英文摘要】 A new stable,high productive strain of Auricularia was bred by inactivated protoplast fusion of the parent strain of Auricularia auricula and Auricularia polytricha Through primary and secondary screening.The Auricularia auricula and Auricularia polytricha protoplast was inactivated with heating(65℃,30min)and UV light (30W,30cm,10min)respectively.The livability of both is about 3 5×10 -7 ~3 2×10 -8 .Under the existing of the inducer of Polyethylene glycol,the parent protoplast fusion is carried out with a recombination freguency of 4 3×10 -5 .
