【中文摘要】 用雙縮脲法、紫外吸收法、考馬斯亮藍(lán)G -2 5 0染色法三種方法對(duì)灰樹花多糖中蛋白質(zhì)含量進(jìn)行測(cè)定 ,并同微量凱氏定氮法進(jìn)行對(duì)比。結(jié)果表明 :在蛋白質(zhì)量濃度 0 1~ 1 0mg/mL范圍內(nèi) ,其測(cè)量誤差分別為雙縮脲法大于 5 0 % ,紫外吸收法大于 30 % ,考馬斯亮藍(lán)G -2 5 0染色法最低可小于 3%??捡R斯亮藍(lán)G - 2 5 0染色法簡單 ,迅速 ,且相對(duì)較為準(zhǔn)確 ,是測(cè)定灰樹花多糖中蛋白質(zhì)含量的有效方法。
【英文摘要】 The Determination methods of residual proteins contained in grifolan such as Biuret Reaction,UV absorption and the dying method with Coomassie Brilliant Fluka G-250 were studied and compared with the Kjeldahl's minor nitrogendetermining method.The results indicated that the determination deviations are more than 50% for Biuret Reaction,more than 30% for UV absorption method,and less than 3% for the dying method with Coomassie Brilliant Fluka G-250 at least while the concentration of proteins ranges from 0 1 to 1 0 mg/ml.The dying method with Coomassie Brilliant Fluka G-250 possesses the properties of simplicity,rapidness and exactness,so it's an effective method of determining the content of proteins contained in grifolan.
