【中文摘要】 香菇(Lentinusedodes)菌株的DNA片段通過克隆轉移到質粒載體BluescriptM13—(pBS)以獲得部分基因文庫。文庫中三個隨機克隆和一個密環(huán)菌中rDNA克隆被用作限制性片段長度多態(tài)性探針。其中三個探針可檢測到多態(tài)性,揭示了32個野生和栽培香菇菌林的遺傳分離。菌林間的相似性用平均鏈鎖聚類方式(UPGMA)聚類,相似性超過60%的分為一組。23個菌株被分成4組。9個菌林與其它菌株相似性均低于60%而獨立存在。從我們的結果看出,香菇的遺傳分離普遍存在于菌株間而與地理位置沒有相關性。RFLP具有研究香菇種內(nèi)遺傳分離的潛力,并可用作遺傳分析的遺傳標記,因此能夠為育種工作和遺傳分析提供科學依據(jù)。
【英文摘要】 EcoR Ⅰ restriction fragment of genemic DNA from the cultivated mushroom,Lentinus edodes,was cloned into the plasmid vector Bluescript MI 3 (pBS). Three randomclones from the genemic library and one rDNA clone of Armillaria melles were used to probefor restriction fragment length polymorphisms(RFLP).Three of the four probes detectedpolymorphisms and were used to demonstrate genetic divergence and detected polymorphismsin 32 wild and cultivated strains of Lentinus mushroom,The similarities between strains werecalculated using average linkage clustering (UPGMA),Based on 60 percent of similarity,23 strains were separated into four groups and anotlier 9 strains were not clustered intoany group due to their less similarity,From our resultS,the genetic divergence occurs amoungstrains but do not have a connection with geographic position,These results suggest thatRFLP have potentiaI to study interspecific genetic divergence in L.edodes and that RFLPcould be served as genetic markers for formal...更多 genetic analysis and thereby provide thefundation for breeding programmes. 還原
