【摘要】 研究了液體發(fā)酵的溫度、起始pH、碳源、氮源和碳氮比對(duì)廣東蟲草菌絲生物量和胞外多糖的影響。當(dāng)發(fā)酵溫度為25℃,培養(yǎng)基起始pH為7.0~9.0,麥芽膏和大豆?jié){粉分別為碳氮源時(shí)菌絲生物量最高;當(dāng)發(fā)酵溫度為28℃,培養(yǎng)基起始pH為5.0~7.0,葡萄糖和酵母膏為碳氮源時(shí)胞外多糖產(chǎn)量最高;以菌絲生物量和胞外多糖的產(chǎn)量為指標(biāo)最優(yōu)的碳氮比為12∶1。正交試驗(yàn)表明:以菌絲生物量為指標(biāo),優(yōu)化的培養(yǎng)基為30g/L葡萄糖,30g/L麥芽膏,8g/L大豆?jié){粉和10g/L酵母膏;以胞外多糖產(chǎn)量為指標(biāo),優(yōu)化的培養(yǎng)基為40g/L葡萄糖,30g/L麥芽膏,10g/L大豆?jié){粉和10g/L酵母膏。
