利用簡單重復序列分子標記分析我國側耳栽培菌株的遺傳多樣性(英文)

【摘要】 采用FIASCO方法,從糙皮側耳(Pleurotus ostreatus)菌株ACCC 50838分離簡單重復序列分子標記(Simple sequence repeat,SSR),所得標記用于分析我國52個側耳(平菇)栽培菌株多樣性,對于SSR標記相同的菌株再進行ISSR PCR分析。結果表明:18個SSR標記在供試菌株中的等位基因的數(shù)量為3~12個,多態(tài)性指數(shù)(PIC)為0.34~0.84;除了2組(ACCC50618,ACCC 50866和ACCC 50915;ACCC 50249和ACCC50476)共5個菌株之外,其他47個菌株都能利用這18個SSR標記進行有效的鑒別。SSR標記相同的的菌株,其ISSR、體細胞不親合性和出菇性狀(溫型、菌蓋顏色)也相同,這些菌株可能是同物異名。利用這18個SSR標記分析52個菌株的遺傳多樣性,相似性指數(shù)為0.755~0.9995。根據(jù)相似性指數(shù)進行聚類分析,52個菌株分成兩個大的類群,分別為糙皮側耳(P.ostreatus)和肺形側耳(P.pulmonarius)。結合這些栽培菌株的農藝性狀和SSR分子標記的聚類分析結果,我國平菇栽培菌株的遺傳...
