【摘要】 應(yīng)用ISSR(inter-simple sequence repeat)分子標(biāo)記技術(shù)對21株不同來源的樺褐孔菌(Inonotusobliquus)菌株進(jìn)行DNA指紋圖譜分析,結(jié)果表明:從60條引物中篩選出11條ISSR引物對樺褐孔菌基因組DNA進(jìn)行了ISSR-PCR擴(kuò)增,共擴(kuò)增出DNA譜帶183條,其中多態(tài)性譜帶169條,多態(tài)位點百分率為92.3%,遺傳相似系數(shù)變化范圍為0.605~0.880。在遺傳相似系數(shù)為0.763時,可將21個樺褐孔菌菌株劃為6大類群,菌株聚類結(jié)果與菌株的地理分布有關(guān)。
【Abstract】 ISSR(inter-simple sequence repeat) molecular markers were used to differentiate 21 Inonotus obliquus wild-type strains originating from different locations worldwide.A total of 183 bands were amplified using 11 ISSR primers(selected from 60 test primers),of which 169 bands were polymorphic.ISSR similarity coefficients ranged from 0.605-0.880,and a dendrogram based on UPGMA(unweighted pair group method with arithmetic average) cluster analysis revealed that the 21 strains clustered into six group...
