【摘要】 首次利用同位素標(biāo)記相對(duì)和絕對(duì)定量(isobaric tags for relative and absolute quantification,iTRAQ)技術(shù)結(jié)合二維液相色譜串聯(lián)質(zhì)譜對(duì)草菇(Volvariella volvacea)同核和異核菌絲蛋白質(zhì)組進(jìn)行研究。采用QExactive質(zhì)譜鑒定并經(jīng)MASCOT軟件搜庫(kù),對(duì)所有鑒定蛋白進(jìn)行主成分分析(principal component analysis,PCA)、層次聚類(hierarchical clustering)分析和GO(gene ontology)注釋,并進(jìn)行生物信息學(xué)分析。結(jié)果共獲得2335個(gè)不同肽段,鑒定到1039個(gè)蛋白,其中1030個(gè)蛋白具有定量信息。在同核菌絲中顯著上調(diào)蛋白83個(gè),下調(diào)蛋白97個(gè),表明iTRAQ標(biāo)記技術(shù)結(jié)合二維液相色譜串聯(lián)質(zhì)譜可有效地分離和鑒定草菇菌絲蛋白質(zhì)組。該研究結(jié)果為更深入全面地研究草菇菌絲蛋白質(zhì)組學(xué)奠定基礎(chǔ)。
【Abstract】 Protein expression in homokaryotic and heterokaryotic mycelia of Volvariella volvacea has been investigated using isobaric tags for relative and absolute quantification(iTRAQ)-coupled 2D LC-MS/MS(two-dimensional liquid chromatography tandem mass spectrometry).A total of 1039 proteins,including 2335 unique peptides,were identified using Q Exactive mass spectrometry and the MASCOT search engine,and 1030 of these had a quantitative dimension.Of the quantitatively different proteins,83 were signific...
