【摘要】 對吉林省延吉市和云南省昆明市松茸子實體的ITS序列進行遺傳多樣性研究。提取兩個地區(qū)松茸子實體的基因組DNA,應用特異引物對其ITS序列進行PCR擴增及測序,并對云南省昆明市松茸樣品進行克隆及測序。之后從GenBank上獲取5個地區(qū)7條松茸ITS序列,并利用MEGA、BLAST、DNAMAN和ClustalX軟件進行對比分析和構(gòu)建系統(tǒng)發(fā)育樹。對云南松茸樣品克隆發(fā)現(xiàn),ITS序列中有A與G、T與C的顛換,并插入1個堿基T。序列對比發(fā)現(xiàn),松茸ITS序列長度在600 bp~604 bp范圍內(nèi),G+C含量在43.0%~44.2%之間,共有41個變異位點,ITS2變異位點數(shù)要多于ITS1的變異位點數(shù)。同時,多個位點存在著轉(zhuǎn)換、顛換、插入和缺失,表明不同產(chǎn)地松茸ITS序列在進化過程中存在差異,為真菌分類、鑒定等研究提供了重要的資料和依據(jù)。
