ERIC技術(shù)在紫木耳親緣關(guān)系鑒定上的應(yīng)用研究
APPLICATION OF ERIC METHOD TO THE AFFINITY AMONG AURICULARIA STRAINS

摘 要:為了研究紫木耳與黑木耳和毛木耳之間的親緣關(guān)系,利用ERIC分子標(biāo)記、Southern雜交技術(shù)和形態(tài)學(xué)分類法,對(duì)紫木耳、黑木耳和毛木耳三者進(jìn)行了詳細(xì)的研究.在相似性系數(shù)為75%的水平上, 由ERIC所得的聚類圖將7個(gè)菌株分為2組,即黑木耳自成一組,毛木耳和紫木耳為一組,兩者相似性系數(shù)為77%.Southern雜交實(shí)驗(yàn)證明毛木耳和紫木耳之間存在較高的同源性.子實(shí)體的形態(tài)學(xué)特征表明紫木耳與毛木耳的親緣關(guān)系較之黑木耳更為密切,同時(shí)進(jìn)一步說(shuō)明ERIC分子標(biāo)記是準(zhǔn)確可信的.本研究探討了紫木耳的分類地位,認(rèn)為在基因組進(jìn)化上,紫木耳與毛木耳的關(guān)系較之黑木耳更為密切.
