大球蓋菇原生質(zhì)體再生及單核化特性的研究

摘 要:大球蓋菇原生質(zhì)體再生條件及單核化特性結(jié)果。原生質(zhì)體再生速度極快,涂布平板3d后肉眼即可見明顯的再生菌落形成,在PGPM再生培養(yǎng)基上再生率為0.97~2.0%,滲穩(wěn)劑種類對(duì)再生率無明顯影響,但可影響再生菌落形態(tài),液體預(yù)培養(yǎng)1~2d,再生率明顯下降;大球蓋菇原生質(zhì)體單核化率高達(dá)77.6%,且再生雙核體和再生單核體在形成再生菌落時(shí)無時(shí)間差,其生長(zhǎng)速度亦無快慢之分,液體預(yù)培養(yǎng)可顯著減少單核化率,再生單核體中存在親本兩種交配型,但二者的比率不為1。

REGENERATION AND MONOKARYOTATION OF PROTOPLASTS OF STROPHARIA RUGOSO-ANNULAT

Abstract:Regeneration and monokaryotation of Stropharia rugoso-annulata protoplasts were investigated. The results showed that the protoplast regeneration was very fast, and the regenerated colonies could be seen by naked eyes after plating 3d. The regeneration rate was 0.97~2.00% on PGPM. The osmotic stabilizers of mannitol, sucrose and magnesium sulphate had no significant effects on regeneration rate, but could affect the morphology of regenerated colonies. Pre-culture of protoplasts in liquid regeneration media for 1~2d at 25℃ could decrease the regeneration and monokaryotation rate. The protoplast monokaryotation rate was 77.7%. The regenerated dikaryons and monokaryons could be formed on the plates simultaneously, and had no difference in mycelium growth rate and colony-forming time. Two kinds of parent mating types could be found among protoplasted monokaryons, but the ratio was not 1:1. It was the first report about the regeneration and monokaryotation characters of Strapharia rugoso-annulata.
